In Iris germanica L., 'G1', 'Adorn' and 'Rococo', induction and proliferation of embryogenic calli were achieved by culture of leaf-base explants on Murashige and Skoog (MS) medium supplemented with 1 mg l−1 2,4-D, 1 mg l−1 kinetin, 200 mg l−1 casein hydrolysate, 250 mg l−1 proline, 30 g l−1 sucrose and 2.5 g l−1 gellan gum. Among these cultivars, however, only in 'G1' could a suspension culture be established using a liquid N6 medium with 1 mg l−1 2,4-D, 1 mg l−1 kinetin, 200 mg l−1 casein hydrolysate, 250 mg l−1 proline and 30 g l−1 sucrose. Murashige and Skoog medium with 1 mg l−1 gibberellic acid (GA3), 30 g l−1 sucrose and 2.5 g l−1 gellan gum was suitable for somatic embryo formation from suspension cells. When the somatic embryos were transferred to solid, growth regulator-free MS medium and subcultured monthly, 36 shoots were obtained from 20 mg suspension cells. Summary Continuous observation of organelles and other cytoplasmic inclusions in the older stretches of living pollen tubes of Iris pseudacorus shows that in the more attentuated parts of the protoplast they move along single, mainly longitudinally oriented fibrils, corresponding to those previously isolated from other species and shown to contain bundles of uniformly polarised actin microfilaments. The traffic associated with each fibril is unidirectional, but organelles move along them independently, sometimes with conspicuously different velocities. Larger columns of cytoplasm passing along the tube are associated with several such fibrils, as revealed in occasional discontinuities and also in columns isolated from the tube in suitable medium without fixation. The dimensions of the individual fibrils suggest that the bundles of actin microfilaments are not likely to be enclosed in a unit membrane corresponding to a tonoplast. If so, the nature of the continuous cavities traversed by numerous fibrils in the older parts of the pollen tube requires reappraisal, since these are more likely to be volumes of attentuated cytoplasm comparable with that of the central cavity of the sieve tube than vacuoles of the normal plant-cell type.  相似文献

1. One explanation of the evolution of sexual cannibalism, the female’s consumption of a male during or following courtship or copulation, is that this behaviour increases the female’s fitness. This study tests the assumption that a single meal increases female reproductive output significantly in the sexually cannibalistic praying mantid Iris oratoria L. 2. In 38 mating trials, seven of the females cannibalised the males. In order to augment the number of females that fed, an additional nine females were each fed one cricket nymph at the end of the mating trial. 3. Three measures of female reproductive output – the occurrence of oviposition, the mass of the first ootheca, and the number of eggs in the first ootheca – increased significantly with female feeding condition, which was a reflection of food consumed before the mating trial. Females that copulated later in the season tended to lay lighter oothecae. 4. The females’ consumption of a meal during the mating trial, either a conspecific male or a cricket, did not influence any measure of reproductive output significantly, although possible effects upon subsequent oothecae cannot be ruled out. 5. If, as the present study suggests, a single meal provides a negligible or delayed benefit to female reproductive output, the evolution of sexual cannibalism might lie in alternative explanations, which include possible fitness benefits to cannibalistic females in the nymphal stage or possible paternity benefits to the cannibalised males.  相似文献

Summary The nerve supply to the iridic melanophores of the rat was studied with the electron microscope. The adrenergic and cholinergic terminals were identified with the aid of 5-hydroxydopamine, which produces dense-cored 400–800 Å synaptic vesicles in adrenergic axon varicosities, whereas the synaptic vesicles of cholinergic axons remain empty. It was found that both adrenergic and cholinergic terminal axons come in close apposition (200–250 Å) with the melanophores. The appositions have the same appearance as synapses in peripheral tissues. It seems likely that the murine iridic melanophores have a double innervation, although its functional significance is obscure.This work has been supported by grants from Lunds Läkarsällskap, the Swedish Medical Research Council (Project no. Sympathetic denervation of the iris muscle produces increases in both the breakdown of phosphatidylinositol 4,5-bisphosphate (PIP2) and in muscle contraction in response to norepinephrine (NE). To shed more light on the biochemical basis underlying this supersensitivity we investigated: the effects of NE on PIP2 breakdown, measured as myo-inositol trisphosphate (IP3) accumulation, and on muscle contraction in normal and denervated rabbit iris dilator; and the effects of denervation on selected biochemical properties of this muscle. The data obtained from these studies can be summarized as follows: The EC50 values (microM) for NE-induced IP3 accumulation in normal and denervated dilators were 14 and 3, respectively. This accumulation of IP3 was blocked by prazosin (1 microM). The EC50 values (microM) for NE-induced contraction for the normal and denervated muscles were 10 and 0.6, respectively. The NE-induced muscle contraction was blocked by prazosin (1 microM). The t1/2 values (s) for IP3 accumulation in normal and denervated muscles were 31 and 11, respectively, and for contraction the values were 19 and 9, respectively. Denervation increased significantly (15-18%) the basal labelling of phosphoinositides from myo-[3H]inositol, but not from 32P or [14C]arachidonic acid. Denervation had little effect on the activities of the enzymes involved in phosphoinositide metabolism. However, the activities of protein kinase C and Ca2+-ATPase increased in the denervated muscle. It is concluded that sympathetic denervation of the iris dilator renders the coupling between alpha1 receptors and PIP2 breakdown into IP3 and 1,2-diacylglycerol (DG) more efficient.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The Iridaceae contains many hundreds of attractive species that have been overlooked for horticultural development. The incredible variation in flower and leaf shape, size and colour suggest this little-tapped resource could offer great opportunities for developing new commercial ornamental products. Micropropagation has increasingly become a valuable tool assisting breeders to release new species and cultivars into the market more rapidly. Here we review the progress made in Iridaceae micropropagation genus by genus, and highlight the potential for future expansion in this field.  相似文献

Both a complex topography and climate change have huge impacts on the distribution and genetic structure of extant species. Due to the lack of relevant molecular research, little is definitively known about the phylogeography of herbaceous plants in East Asia. Here we investigate the genetic diversity, population structure and historical population dynamics of Iris dichotoma Pallas, a widespread perennial herbaceous species in northeastern and northern China. Twenty-nine populations, totalling 297 individuals, were sampled throughout the Chinese distributional range of I. dichotoma. The combined sequences of six chloroplast DNA fragments (petA-psbE, rps18-clpp, psbJ-petA, trnD-trnT, rps16 and ndhA) were used to identify 13 haplotypes, of which six were private ones restricted in a single population. Genetic differentiation among I. dichotoma populations, enabled us to infer potential refugia during the glacial period in the Yinshan Mountains–Yanshan Mountains, where high levels of haplotype and nucleotide diversity were detected. The results of a neutral test and mismatch distribution analysis both indicated that I. dichotoma underwent a recent population expansion. In East Asia, postglacial environmental and climatic changes appear to have promoted genetic diversification not only in better-studied woody species, but also in herbaceous ones like I. dichotoma. Future studies of more herbaceous plant species are needed to obtain better insight into how modern temperate biodiversity has developed in East Asia.  相似文献

Premise of the study: Microsatellite primers were developed in Iris ensata (Iridaceae) to provide polymorphic markers for further studies into population genetics. • Methods and Results: Thirteen polymorphic microsatellite loci were isolated from I. ensata. These loci were successfully amplified in two natural populations of I. ensata from eastern China (Longwangshan, Zhejiang Province) and northeastern China (Jinchuan, Jilin Province). There was no significant linkage disequilibrium found for any pair of loci. These loci contained between two and 12 alleles per locus across all 48 individuals of I. ensata. The number of alleles per locus varied from two to 10 at the population level and the observed and expected heterozygosities ranged from 0.167 to 0.958 and from 0.284 to 0.853, respectively. • Conclusions: These loci showed high levels of polymorphism and could be used to study the population genetic structure, genetic relationships, and phylogeography of I. ensata.  相似文献

The acceptability of dietary allopurinol to German cockroaches, Blattella germanica (L.), was assessed. In diet choice tests between rat chow with or without 0.1% w/w allopurinol there were no significant differences in nymphal development, feeding duration, visits or consumption. The LT50 of cockroaches fed a choice of diets was 3 weeks greater (7.88) than those fed non-choice a 0.1% allopurinol diet. Female cockroaches provided a choice of diets aborted a significantly greater percentage of their oothecae (98.6%) than those fed the untreated diet (1.7%). Choice arena tests of 2% allopurinol in rat chow significantly reduced cockroach populations compared with untreated controls. After 6 weeks, populations were reduced by approximately 50%, and 97% after 14 weeks. These results indicate that allopurinol is acceptable to B. germanica as a dietary supplement which could be used in baits for cockroach control.  相似文献
